Key Regulator of Intestinal Homeostasis Identified

SP140, an epigenetic reader protein mutated in a number of autoimmune disorders, is essential for macrophage function and preventing intestinal inflammation, scientists show.

Researchers are only beginning to understand the roles of the hundreds of proteins involved in reading, writing, and erasing the epigenome. One of the epigenetic regulators, SP140, which is mutated in a number autoimmune disorders, including Crohn’s disease and multiple sclerosis, is also essential to macrophage function and intestinal homeostasis in both humans and mice, scientists reported today (March 3) in Science Immunology.

“Many immune-mediated disorders are driven by a combination of genetic susceptibility as well as environmental influences [so] epigenetics is a suitable critical juncture between those two aspects of the disease,” said coauthor Kate Jeffrey, a researcher investigating the epigenetic control of innate immunity at Massachusetts General Hospital.

An estimated 230,000 Australians have chronic hepatitis C, and a quarter of cases are undiagnosed.

Hepatitis C inflames the liver and unlike the A and B viruses there is no vaccine available. Pathology is important for diagnosing the virus.

Many people with hepatitis C may not experience symptoms, but left untreated the disease can cause cirrhosis (scarring of the liver), which in a small number of cases can lead to liver cancer.

Serum Albumin Levels Associated with Mortality in Hemodialysis Patients

Serum albumin is commonly used as a proxy for nutritional status, as well as a marker of inflammation and a low serum albumin concentration is not only indicative of protein energy wasting in dialysis patients, but it is also a powerful predictor of the mortality risk in this population.

Several clinical conditions are associated with low serum albumin levels in dialysis patients, including infectious and inflammatory diseases, fluid overload, inadequate dialysis, severe co-morbidity, and taste change. Therefore, regular monitoring of serum albumin levels is useful for predicting outcomes in dialysis patients.

A new study reveals an important molecular clue about how inflammation may lead to prostate enlargement or benign prostatic hyperplasia. The researchers believe their discovery may also lead to ways of overcoming resistance to androgen-targeted treatment for the condition.

Enlarged prostate or benign prostatic hyperplasia (BPH) is a common condition in older men where the prostate gland is enlarged and not cancerous.

As the prostate gland enlarges, it presses against and pinches the urethra, the tube that allows urine to leave the bladder. Also, the bladder wall gradually thickens and the bladder may get weaker and lose the ability to empty completely.


Enlarged prostate or benign prostatic hyperplasia (BPH) is a common condition in older men.

Cracking the Enigma of Fatty Liver Disease

Nonalcoholic fatty liver disease (NAFLD) affects one out of four people in the world, making it the most prevalent of all liver diseases and a major public health problem. The disease is defined by abnormally increased fat deposition in liver cells, which can progress with the addition of inflammation and cell damage to nonalcoholic steatohepatitis (NASH), leading to progressive stages of fibrosis and, finally, cirrhosis. Early NASH is characterized by fat and inflammation associated with minimal amounts of fibrosis, and late NASH is associated with advanced fibrosis or cirrhosis. NAFLD is most closely associated with obesity and diabetes, although there are likely other genetic, nutritional, and environmental factors involved in the pathogenesis.

Although there are no approved pharmacologic treatments for NAFLD, NASH, or cirrhosis, there are over a dozen different companies researching possible treatments. However, the development of drug therapies for the NAFLD spectrum of disease is hampered by a number of challenges:
  • the chronic nature of the disorder and ultimately low morbidity and mortality,
  • the impact of weight loss on the disease,
  • a complex and poorly defined pathophysiology,
  • the lack of easily administered diagnostic testing, and
  • the evolving status of regulatory endpoints.

The results of multiple clinical trials over the next few years will clarify potential therapies and target

All You Need to Know About Nephritis

To understand kidney problems such as nephritis, it's helpful to start with some background on what the kidneys are, and what they do.

The kidneys are two bean-shaped, fist-sized organs found just under the ribs on the left and right sides of the spine. They remove impurities and extra water from the blood, filtering 120-150 quarts of blood a day, according to the National Institute of Diabetes and Digestive and Kidney Diseases.

Each kidney consists of thousands of structures called nephrons, where the actual blood filtering takes place. In the nephron, a two-step cleaning process separates what the body needs to keep from what it can get rid of.

A filter called the glomerulus catches blood cells and protein, sending water and waste to a second filter, called a tubule. The tubule captures minerals. After that, what remains leaves the body as urine.


Nephritis can lead to kidney failure if not treated. There are various ways of preventing kidney damage
if someone has symptoms of nephritis. Monitoring blood pressure is important if kidney problems
develop.

Bacterial Pneumonia - Get the Facts

Bacterial pneumonia is an inflammation of the lungs due to some form of bacteria. There are different types of bacteria that may lead to the infection.

The lungs are made up of different sections or lobes. There are three lobes on the right and two on the left. Bacterial pneumonia can affect both lungs, one lung, or even just one section of a lung.

The lobes of the lungs are made up of small air sacs called alveoli. Normally, the air sacs fill with air. Oxygen is inhaled and carbon dioxide in exhaled.

When a person develops pneumonia, the air sacs become inflamed, which can cause them to fill with fluid. If the air sacs are filled with fluid instead of air, it can make breathing difficult. In some cases, the lungs may not get enough oxygen.


Bacteria that cause pneumonia are spread through coughing. Doctors will carry out a chest X-ray to
assess fluid buildup in the lungs. An annual flu vaccine can help prevent pneumonia.

may be needed to treat a case of appendicitis.

Identifying and Treating Inflammation

There are three types of inflammation: acute, chronic and life-threatening.

When basketball star Steph Curry slipped and injured his knee in late April, he reacted by tearing up on the sideline, while fans and media reacted by wondering how many games he would miss. His body? It reacted by inflaming around the outside of the knee, protecting the area as it recovered from a medial collateral ligament sprain.

The word inflammation can conjure up visuals of a swollen, red and throbbing knee or, thanks to some TV ads, a lit match doused by a medicated pad. Conventional public wisdom frames inflammation as being adverse, painful and harmful – something most people want to resolve quickly with medication.


Treat acute inflammation by managing it, not trying to cure it.

Breast milk hormones found to impact bacterial development in infants' guts

Intestinal microbiome of children born to obese mothers significantly different from those born to mothers of healthy weight

A new study finds that hormones in breast milk may impact the development of healthy bacteria in infants' guts, potentially protecting them from intestinal inflammation, obesity and other diseases later in life.

The study, published Monday in the American Journal of Clinical Nutrition, examines the role of human milk hormones in the development of infants' microbiome, a bacterial ecosystem in the digestive system that contributes to multiple facets of health.

"This is the first study of its kind to suggest that hormones in human milk may play an important role in shaping a healthy infant microbiome," said Bridget Young, co-first author and assistant professor of pediatric nutrition at CU Anschutz. "We've known for a long time that breast milk contributes to infant intestinal maturation and healthy growth. This study suggests that hormones in milk may be partly responsible for this positive impact through interactions with the infant's developing microbiome."

Researchers found that levels of insulin and leptin in the breast milk were positively associated with greater microbial diversity and families of bacteria in the infants' stool.

Immune cells self-healing brain after stroke

After a stroke, there is inflammation in the damaged part of the brain. Until now, the inflammation has been seen as a negative consequence that needs to be abolished as soon as possible. But, as it turns out, there are also some positive sides to the inflammation, and it can actually help the brain to self-repair.

"This is in total contrast to our previous beliefs", says Professor Zaal Kokaia from Lund University in Sweden.

Zaal Kokaia, together with Professor of Neurology Olle Lindvall, runs a research group at the Lund Stem Cell Center that, in collaboration with colleagues at the Weizmann Institute in Israel, is responsible for these findings. Hopefully, these new data will lead to new ways of treating stroke in the future. The study was recently published in the Journal of Neuroscience.

When stroke occurs, the nerve cells in the damaged area of the brain die, causing an inflammation that attracts cells from the immune system. Among them you find monocytes—a type of white blood cells produced in the bone marrow.

Atherosclerosis, Rheumatoid Arthritis and Inflammation.

Rheumatoid arthritis (RA) has long been associated with increased cardiovascular risk, but despite substantial improvements in disease management, mortality remains high. Atherosclerosis is more prevalent in RA than in the general population, and atherosclerotic lesions progress at a faster rate and might be more prone to rupture, causing clinical events. Cells and cytokines implicated in RA pathogenesis are also involved in the development and progression of atherosclerosis, which is generally recognized as an inflammatory condition. The two diseases also share genetic and environmental risk factors, which suggests that patients who develop RA might also be predisposed to developing cardiovascular disease. In RA, inflammation and atherosclerosis are closely linked. Inflammation mediates its effects on atherosclerosis both through modulation of traditional risk factors and by directly affecting the vessel wall. Treatments such as TNF inhibitors might have a beneficial effect on cardiovascular risk. However, whether this benefit is attributable to effective control of inflammation or whether targeting specific cytokines, implicated in atherosclerosis, provides additional risk reduction is unclear. Further knowledge of the predictors of cardiovascular risk, the effects of early control of inflammation and of drug-specific effects are likely to improve the recognition and management of cardiovascular risk in patients with RA.

Rheumatoid arthritis (RA) is associated with a significantly increased risk of cardiovascular mortality, accounted for mainly by increased atherosclerotic disease.1, 2 Although the prevalence of some traditional cardiovascular risk factors is increased in RA, adjustment for these factors does not fully account for the heightened risk, suggesting that RA itself is an independent risk factor for cardiovascular disease (CVD).3 The prevalence of atherosclerosis is increased in RA, even in early disease,4 and chronic inflammation is thought to promote atherosclerosis both by modulation of traditional risk factors and also possibly by direct biological effects on the artery. In this article, we discuss the potential mechanisms that might accelerate atherosclerosis in RA, with a particular focus on inflammation.
